The new GRASS 5.1 which is work-in-progress will be able to
do that:

Since GRASS5.1 is highly experimental I cannot recommend to use it
now, but you can perform vector network analysis now (since one week).
GRASS 5.1 also reads directly from SHAPE file, you just register it
into the GRASS location with a script.

Watch out on above page for "vector network analysis".

Of course the development team would be glad to welcome more
programmers for GRASS5.1!
