Many Free Software products can do this in principle.
E.g. gpsdrive, tkgeomap, openmap, thuban.
You might need to adapt them to your needs.

This is also relatively easy.

This one would require quality street level vector data
and a routing algorithm.  Thus is it a complicated task.

E.g.  GRASS (5.7), BBBike and Hoko could do it the routing part.
If you have the data.

It depends on the wanted integration.
If all results go into a PostGIS database,
any other client could just read the results.
Mapserver is a good choice to create bitmap results and so on.

Your requirements are not precise enough 
to recommend the best software. 
And it probably will be work to find the right one.

My examples above show that there are many Free Software GIS products
that have the principal technical abilities to do the job.
